  Working Principles
Click to ViewHPLC works on the principle that some molecules take longer than others to pass through a chromatography column. This depends on the affinity of the molecule with the mobile phase (liquid or gas) and the stationary phase (solid or liquid). The ones that have more affinity with the stationary phase take longer to pass through and vice versa. Reversed-phase chromatography is a technique using alkyl chains covalently bonded to the stationary phase particles in order to create a hydrophobic stationary phase, which has a stronger affinity for hydrophobic or less polar compounds. Reversed-phase chromatography employs a polar (aqueous) mobile phase. As a result, hydrophobic molecules in the polar mobile phase tend to adsorb to the hydrophobic stationary phase, and hydrophilic molecules in the mobile phase will pass through the column and are eluted first. Hydrophobic molecules can be eluted from the column by decreasing the polarity of the mobile phase using an organic (non-polar) solvent, which reduces hydrophobic interactions. The more hydrophobic the molecule, the more strongly it will bind to the stationary phase, and the higher the concentration of organic solvent that will be required to elute the molecule.

Reversed-phase HPLC is very widely applicable for small molecule analyses (M.Wt. < 2000 Da) like metabolites. Reversed-phase HPLC is also a good choice for peptide and protein separations when short chain alkyl stationary phases are used. Here, analysis of compounds with a molecular weight above 2000 Daltons is possible. Samples should be processed in HPLC grade solvents and filtered through 0.45µm filter and stored in suitable vials.
