Click to ViewFirst described by Solomon Berson and Rosalyn Yalow in 1960, radioimmunoassay (RIA) is a highly specific and sensitive assay for detection and quantification of radioactive radioisotopes conjugated with antigens or antibodies. The radiolabelled antigen and antibody complex when used in a reaction with unlabelled antigen or antibody can be used to quantitative the unknown amounts of respective unlabelled antigen/antibody the test biological sample in a reaction. Such classical reactions follow the principle of competitive binding.

Click to View•Competitive binding assay in which fixed amount of antibody and labeled antigens reacts in presence of known (standard) or unknown (sample) amount of antigen •Measures very low concentration of analytes in the biological fluids which is based on a class of saturation analysis. 125-I labels are usually applied although other isotopes such as C14 and H3 have also been used. Usually, high specific activity radiolabeled (125-I) antigen is prepared by iodination of the pure antigen on its tyrosine residue(s) by chloramine-T or peroxidase methods and then separating the radio-labeled antigen from free-isotope by gel-filtration or HPLC.
