  Working Principles
Click to ViewProtein content in food samples is estimated by determining their nitrogen content and multiplying it with factor of 6.25. The sample is digested by heating it with concentrated sulphuric acid in the presence of salt and metal catalyst. Digestion converts nitrogen in all organic molecules to ammonia and is trapped in the form of ammonium sulphate. Ammonia is steam distilled and trapped in acid. Ammonia concentration is then determined by back titration with standardized acid.
