Click to ViewThe Perkin Elmer AutoSystem XL Gas Chromatograph (GC) is a partially automated gas chromatograph. All instrument functions are set up and monitored through a PC for analyses of fatty acid methyl esters (FAMEs). FAMEs will be analyzed by an Perkin-Elmer AutoSystem XL gas chromatograph (Perkin-Elmer) equipped with an Elite-5 (cross-bond 5% diphenyl polsiloxane-95% dimethyl polsiloxane) capillary column (30 m X 0.53 mm i.d.; Supelco, Bellfonte, PA) using a flame ionization detector (FID). FAMEs are identified by comparison of retention times with the known standards (37-component FAME mix; Supelco).

  Working Principles
Click to ViewThe sample solution needs to be injected into the instrument enters a gas stream which transports the sample into a separation tube known as the "column" (helium or nitrogen is used as the so-called carrier gas). The various components are separated inside the column. For fatty acid analysis, the FAME samples need to be extracted by the cold extraction method using a chloroform-methanol mixture in a 2:1 ratio (Folch et al., 1957). After isolating the lipid phase, the solvent is evaporated. Two millilitre sample needs to be refluxed with 5 ml of 0.5% alcoholic potassium hydroxide solution for 30 min and then refluxed with 6 ml of BF3 -MeOH (Sigma-Aldrich, Bangalore, India) for an additional 5 min. Refluxing needs to be carried out under nitrogen. The dry fatty acid methyl esters in the flask need to be extracted with petroleum ether (10 ml x 3) quantitatively. The extract needs to be washed with 25 ml double distilled water three times and filtered over anhydrous sodium sulphate (10 g) to remove any moisture. The solvent needs to be evaporated under a stream of nitrogen gas. Two microlitre each of the prepared fatty acid methyl ester samples (FAME) need to be injected into an Auto-System XL Gas chromatograph (Perkin Elmer, Waltham, MA, USA) according to Morrison and Smith (1964).

Analysis of fatty acid profile of fish tissue.The samples should be extracted, and fatty acid methyl ester samples (FAME) will be accepted for injection only. The methodology under the head of “working principles” may be followed. No sample processing will be done at this facility. MS-DS (Material Safety Data Sheet) should be given along with samples to ensure that there is no toxic sample being given. Samples should not be toxic or hazardous. Samples will not be accepted unless accompanied by MS-DS. Only machine is available for use, client has to bring his/her own plastic ware etc, which are required. The facility is available during working hours of ICAR-CMFRI.
