Click to ViewThe FOSS KJELTEC 2300 Protein Analyzer is a semi-automatic system comprising of a digestion unit and fully automatic distillation and iteration units. A total of 20 samples can be analyzed in a row. Analysis time: 3-5 minutes per sample.

  Working Principles
Click to ViewWorking Principle: The method consists of three basic steps: (i) digestion of the sample (ii) distillation and (iii) quantification of the ammonia by titration. The sample is heated with concentrated sulphuric acid, which decomposes the organic substance by oxidation to liberate the reduced nitrogen as ammonium sulphate. In this step potassium sulphate and copper sulphate are added as catalysts. Chemical decomposition of the sample is complete when the initially dark-coloured medium becomes clear and colourless. The solution is then distilled with 40 % sodium hydroxide, which converts the ammonium salt to ammonia which is trapped into a solution of 4 % boric acid. The amount of ammonia present, and thus the amount of nitrogen present in the sample, is determined by titration with a standard solution. Digestion: Sample + Con. H2SO4 → (NH4)2SO4(aq) + CO2(g) + SO2(g) + H2O(g) Distillation: (NH4)2SO4(aq) + 2NaOH → Na2SO4(aq) + 2H2O(l) + 2NH3(g) Capture of ammonia: NH3 + H3BO3 (boric acid) → NH4+ + H2BO3- Titration: H2BO3- + H+ → H3BO3 Applications: Estimation of crude protein content of fish tissues, raw materials and finished products in feeds
