  Working Principles
Click to ViewThe spectrophotometer consists of five parts: 1) Halogen or deuterium lamps to supply the light; 2) A Mono-chromator to isolate the wavelength of interest and eliminate the unwanted second order radiations; 3) A Sample compartment to accommodate the sample solution; 4) A detector to receive the transmitted light and convert it to an electrical signal; and 5) A digital display to indicate absorbance or transmittance. In this spectrophotometer, light from the lamp is focused on the entrance slit of the mono-chromator where the collimating mirror directs the beam onto grating. The grating disperses the light beam to produce the spectrum, a portion of which is focused on the exit slit of the mono-chromator by collimating mirror. From here the beam is passed to a sample compartment through one of the filters, which helps to eliminate unwanted second order radiation from the diffraction grating. Upon leaving the sample compartment, the beam is passed to the silicon photodiode detector and causes the detector to produce an electrical signal that is displayed on the digital display.

Spectrophotometers offer a variety of uses and applications in agriculture and soil quality analysis. The use of spectrophotometers spans various scientific fields, such as chemistry, biochemistry, physiology and biology. Spectrophotometer is often used in measurements of enzyme activities, determinations of protein concentrations, determinations of enzymatic kinetic constants and soil and plant nutrients. Ultimately, a spectrophotometer is able to determine, depending on the control or calibration, what substances are present in a target and exactly how much through calculations of observed wavelengths. Portability, durability, and rapid speed of evaluation are all valuable characteristics of how color technology is making its mark in this field.For extracting the chemical compound/nutrient in coloured solution/turbid solution standard method should be followed. Do not use any method which encompasses hazardous chemicals in their protocol. Sample solution should be filtered by using proper number of filter paper to avoid further problems during handling.
